New Dosing Recommendations for Trilostane
in Cushing's Disease

Trilostane is actually not a new medication in the treatment of Cushing's disease. We have been using it for four to five years. However, what is new is the change in dosing schedules. Previously most endocrinologists recommended a once-daily dosing. At this point, the dosing recommendations is twice daily with the dose being much smaller. This change reflects further research into the effectiveness and blood levels of Trilostane using it once or twice daily. Research has shown repeatedly, that blood levels of Trilostane are higher when the medication is given twice daily.

Trilostane is a safe and very effective medication for the treatment of Cushing's Disease (pituitary hyperadrenocorticsm). The drug only inhibits one of the final steps in making cortisol. There are no untoward effects on the adrenal glands as were oftentimes seen using the older treatment (mitotane).
